Paper: Enebo et al., The Lancet (2021) — a Phase 1b/2a trial of cagrilintide co-administered with semaglutide in adults with overweight or obesity.
What it tested: tolerability and pharmacodynamics of the combination (the early read on whether pairing an amylin analog with a GLP-1 makes sense).
What it found: the combination was tolerable and produced effects consistent with added weight reduction — enough to justify advancing the program.
Caveats: early-phase, modest size, short duration. It's a foundation result, not a confirmatory one — which is why cagrilintide is graded B and the later Phase 3 readouts matter so much.
