Kisspeptin-10 is a 10-amino-acid C-terminal fragment of the KISS1 gene product, a peptide family that includes kisspeptin-54, kisspeptin-14, kisspeptin-13, and kisspeptin-10. All variants share the same C-terminal sequence that binds the kisspeptin receptor (KISS1R, also called GPR54).
Kisspeptin signaling at the level of hypothalamic GnRH neurons is now understood to be the dominant upstream regulator of the hypothalamic-pituitary-gonadal axis. The discovery that loss-of-function mutations in KISS1R cause hypogonadotropic hypogonadism in humans (reported in 2003) reframed reproductive endocrinology and made kisspeptin a focal point for both basic and clinical research.
